  •        新陳代謝是機體生命活動的基礎,影響細胞的各個方面。生長、增殖、分化或凋亡等不同的細胞過程,存在著顯著不同的代謝網絡[1,4]。細胞內的新陳代謝除了受信號通路等多種機制調控外,也可作為信號分子,或獨立調控生物學過程[8]。最近研究表明,新陳代謝參與調控表觀遺傳[6,7]、自噬[3,5]、凋亡、壞死[9]等多種細胞生命活動。代謝異常是引起肥胖、糖尿病及腫瘤等多種疾病的重要因素(圖1)。腫瘤被認為是一種代謝疾病,代謝改變是癌癥發生的顯著標志之一。癌細胞進行代謝重編程,以滿足腫瘤轉化、起始、發展、侵襲與遷移等不同階段以及不同營養環境下的代謝需求[1,8](圖2)。系統地分析疾病的代謝改變,對尋找有效的代謝標記物與疾病治療靶點非常必需。
          Arraystar公司新推出的NuRNA? Human Central Metabolism PCR Array,可同時檢測373個編碼重要代謝酶與代謝物轉運蛋白的轉錄本,是進行代謝研究必不可少的工具。該芯片檢測的代謝通路與代謝物轉運系統包括:葡萄糖轉運蛋白,糖酵解、TCA循環、脂類合成、乳酸合成及轉運、糖異生途徑、糖原代謝、氨基己糖代謝、磷酸戊糖旁路、一碳單位代謝、谷氨酰胺轉運及代謝、氧化還原平衡與GSH合成、脂肪酸氧化、乙酸代謝、核苷酸代謝等。研究不同生物學過程的代謝特征,尋找生理病理狀態下的代謝改變,將有助于深入理解細胞生命活動,并為代謝疾病的治療提供新的思路。
